They have met on just one previous occasion at the Keepmoat Stadium when they met in a League One clash in the 2007/08 season.
After a tight goalless first half the game sprung to life in the second half when Richie Wellens fired Rovers ahead.
Jabo Ibehre quickly restored parity and Orient went ahead halfway through the half through Stephen Purches.
Jason Price equalised within a minute for Rovers and goals in the final ten minutes from James Hayter and a second for Wellens secured a 4-2 Rovers win.
